Characteristics of the Cytokine Response in Patients with Differenx Forms of Juvenile Idiopathic Arthritis
CHARAKTERISTIKA CYTOKÍNOVEJ ODPOVEDE U PACIENTOV S ROZNYMI FORMAMI JUVENILNEJ IDIOPATICKEJ ARTRITÍDY

Narušenie dynamickej rovnováhy medu prvým a druhým typom cytokínovej odpovede s prevahou cytokínov prvého typu hrá úlohu v patogenéze mnohých autoimunitných chorób vrátane juvenilnej idiopatickej artritidy (JIA). Ciel'om práce bolo analyzovat cytokínový profil lymfocytov periférnej krvi u pacientov s JIA a vyhodnotit vzájomný pomer medu lymfocytmi T produkujúcimi cytokíny 1. a 2. typu pri jednotlivých formách tejto choroby.Metódy a výsledky. Vyšetrili sme súbor 42 detí vo veku od 6-16 rokov, 32 chorých s JIA a 10 detí s neimunologickými chorobami, ktoré tvorili kontroloú skupinu. Expresiu intracelulárnych cytokínov INF-y a IL-4 sme stanovovali v aktivovaných lymfocytoch T podl'a odporúčaného pracovného protokolu (FastImmune Cytokine system, BD). Na analýzu vzoriek sme použili prietokový cytometer FACScan (BD). Vo vyšetrenom súbore pacientov sme nezistili rozdiel medu zastúpením CD3+INFy+ Ly u pacientov s oligoartikulárnou a polyartikulárnou formou JIA (19,3±8,65 vs. 19,2±9,7 %), ktoré bolo rovnaké ako v kontrolnej skupine (19,5±5,85 %). Podobne sme nedokázali rozdiel v zastúpení CD3+IL4+ Ly u chorých s oligoartikulárnou formou JIA v porovnaní s kontrolou (2,4±0,9 vs. 2,8±0,45 %). Background.
Impairment of the dynamic balance between the first and second type of cytokine response with the prevalence of cytokines of the řust type plays and important role in the pathogenesis of several autoimmune diseases, including the juvenile idiopathic arthritis (JIA). The aim of our work was to analyse cytokine profile in lymphocytes in the peripheral blond of patients with JIA, and to evaluate the mutual radon between T lymphocytes producing cytokines of the řust and second type in different forms of the disease.Methods and Results. We studied a group of 42 children aged 6 to 16 years, 32 of them were patients with JIA, 10 children with non-autoimmune diseases represented the control group. Expression of intracellular cytokines INF-y and IL-4 were assayed in activated T lymphocytes using recommended protocol (FastImmune Cytokine system, BD). For the sample analysis, flow cytometer FACScan (BD) was used. No differences in incidence of CD3+INF-y + Ly was found between patients with oligoarticular and polyarticular form of JIA (19.3±8.65 vs. 19.2±9.7 %), and values were equal to that of the control group (19.5±5.85 %). Similarly, no difference was found in incidence of CD3+INF4+ Ly between the JIA patients and controls (2.4±0.9 vs. 2.8±0.45 %).
