GENOTYPIZACE HLAVNÍHO HISTOKOMPATIBILNÍHO SYSTÉMU HLAPRO POTŘEBY TRANSPLANTACE HEMATOPOETICKÝCHKMENOVÝCH BUNĚK

Transplantace kostní dřeně či periferních kmenových buněk je součástí standardních protokolů v léčení hematologickýchmalignit i metabolických poruch a imunodeficiencí. Z hlediska kompatibility dárce a příjemce HSC je rozhodující shoda v pětiHLA lokusech: HLA-A,-B,-C,-DRB1,-DQB1. Míra shody mezi pacientem a dárcem v HLA systému, který je nejpolymorfnějšímgenetickým systémem člověka, je spolehlivě detegovatelná pouze na úrovni DNA. Naše pracoviště provádí určování HLAkompatibility pro potřeby Ústavu hematologie a krevní transfuze a 2. dětské kliniky FN Motol. Ročně se jedná asi o 200vyšetřených osob.

Overview

Transplantation of bone marrow or peripheral stem cells is part of the standard protocols in the treatment of haematologicalmalignities and metabolic disorders and immunodeficiencies. From the aspect of compatibility of the donor and recipient ofHCS agreement in five HLA locuses is decisive: HLA-A, B-, C-, DRB1-, DQB1-. The extent of agreement between the patient anddonor in the HLA system which is the most polymorphous genetic system of man is reliably detectable only at the DNA level.Our department assesses the HLA compatibility for the needs of the Institute of Haematology and Blood Transfusion and theSecond Paediatric Clinic of the Motol Faculty Hospital. Every year some 200 subjects are examined.
